NAD+ is an obligate redox cofactor (the NAD+/NADH couple) and the consumed substrate of NAD+-dependent enzymes: sirtuins (SIRT1–7 deacylases), PARPs (DNA-repair signaling), and CD38. Preclinical literature examines cellular NAD+ availability in relation to mitochondrial oxidative phosphorylation, SIRT1/SIRT3 activity, genome maintenance, and the documented decline of tissue NAD+ pools with age.
